An immediate response is the activation of (PKR)-like-ER kinase (PERK) that leads to phosphorylationof eukaryotic initiation factor-2 alpha, thereby attenuating protein synthesis.We demonstrated that calcineurin-A/B(CN-A/B),a heterodimeric Ca2+ phosphatase, associates with PERK, increasingits auto-phosphorylation and significantly enhancing inhibition of protein translationand cell viability in Xenopusoocytes.                  Here, we report that the level of CNAb/B, the astrocytes specific CN isoform, as well as PERKphosphorylation were significantlyincreased after Oxygen and Glucose Deprivation (OGD) treatment. Furthermore, overexpressionof CNAb increased theviability of wild-type astrocytes exposed to OGD but not in astrocytes null for PERK(-/-). Kinase assays with recombinantproteins demonstrated that PERK autophosphorylation was increased in presenceof CNAb/B but not in the presence of only regulatorysubunit B.  Finally, we observed thatlight- or chemical-induced heterodimerization of PERK and CNAb/B, leads to increase PERK phosphorylation inastrocytes. It was markedly enhances by thapsigargin treatment, suggesting thatCa2+ release from the ER further enhanced the CN-PERK interaction. Taken together, our findings indicate that the protective role of CN-PERKinteraction observed in Xenopusoocytes can be extended to astrocytes stressed by OGD.  Funded in part by NIHR21-NS085732, andPID-2010.
