Motor behavior changes in a murine model of encephalopathy produced by Shiga Toxin type 2 (Stx2) from enterohemorrhagic Escherichia coli. Interaction between different diets.

Stx2 may damage the CNS independently or concomitantly with Hemolytic Uremic Syndrome (HUS). Different types of diets may play a critical role on Stx2 pathogenicity, including neurological motor dysfunction that may occur following intoxication with Stx2. Actually there are no reports regarding this issue. However, motor neurological alterations in patients frequently occur. Such dysfunctions may be caused by alteration of the neurological motor system. Thus, the aim of this study was to determine by a behavioral test whether this motor neurological alteration occurs after intravenous administration of a sub-lethal dose of Stx2 in 4 different diet groups. Male NIH mice were subjected to 4 different diet groups (normal diet-control-, high fat diet, high carbohydrates diet, and a combination of the last two diets) for 16 days after the weaning. They were then injected intravenously (i.v.) with vehicle (vehicle group) or 1ηg of Stx2 per mouse (sub-lethal dose). The swimming test was performed before and after 4 days of their respective i.v. treatment, and 2 variables were analyzed, the time of swimming and the number of hindlimbs plus forelimbs kicks. A two-way analysis of variance (ANOVA) test, following by Bonferroni post hoc test were done to analyze the data.
