Resumen: Cytokines in the male reproductive tract play an immunomodulatory role and several evidences indicate that an imbalance of cytokine production can affect fertility. In addition, immunity against sperm antigens has been related to infertility. We have previously demonstrated that the proacrosin/acrosin system is immunogenic in Balb/c and CF1 male mice and antibodies significantly correlate with decreased fertility. The aim of this study was to evaluate the immune profile induced in male genital tract of mice immunized against proacrosin. CF1 male mice received 4 doses of 50µg of a plasmid containing the cDNA of human proacrosin (pSF2-Acro, n=12), and the control group, the empty plasmid (pSF2, n=8), every 3 weeks. Total anti-proacrosin IgG, IgG1 and IgG2a antibody levels, also interleukin (IL)-4 and IFN-g, were assessed by ELISA in serum and in seminal vesicle fluid (SVF). RNA from testis and seminal vesicles was obtained to test expression levels of cytokines by real time PCR, using cyclophilin as housekeeping gene.
