Eco-friendly chitosan-based nanoparticles for beta vulgaris extract encapsulation. Development, characterization, antioxidant efficiency, biocompatibility and cytoprotective studies in hTERT-RPE cells

Phytochemicals distinctly decrease the risk of diverse degenerative and chronic diseases. Betanin (BET) is a polyphenol that displays a great potential to mitigate oxidative stress and inflammation. Still, this pigment is labile to harsh environmental conditions. Nanotechnology emerges as a possible solution to improve phytochemicals´ limitations. Hence, the main object of this study was to develop and characterize nanoformulations (NF) based on a novel chitosan (CS) crosslinked with tripolyphosphate (TPP) to encapsulate BET from red beetroot extracts (RBE). Results: the BET fractions present in the RBE were analyzed by thin layer chromatography and UV-visible spectroscopy and compared with standard commercial BET. Spectroscopy analysis showed that BET has a prominent presence in the purified fractions. We determined by dynamic light scattering analysis that the mean size of particles present in the NFs loaded with BET was determined based on the QS:TPP ratio. The values were: NF-2:1 (130nm), NF-3:1 (157nm), NF-4:1 (170nm), NF-5:1 (182nm). All NFs presented a monodispersity profile with a desire low PdI (NP: 0.16; NP-BET: 0.22). BET percentage of encapsulation efficiency (EE) were NF-2:1 (15%), NF-3:1 (40%), NF-4:1 (41%), NF-5:1 (42%).
