Between X-ray protein crystallography and cryo-electron microscopy: model building at low resolution

The first 3D structure available for a high-molecular weight glutamate dehydrogenase (H-GDH) is reported. The mycobacterial H-GDH is a 7 kDa homo-tetramer, with inter-subunit surfaces provided by a multi-modular N-terminal domain containing ACT-like and PAS modules and a C-terminal helical domain flanking the catalytic core. Two protein conformations were found with different inter-subunit geometries, providing clues about the structural bases of allosteric regulation.
