Previously, we demonstrated that
Progesterone (P4) added to the coeliac ganglion (CG), via the
superior ovarian nerve (SON), stimulated the ovarian P4 release, decreased the luteal
expression of 20α-HSD,
P4 degradation enzyme and the FasL expression, regulator factor of the way
extrinsic of apoptosis. The aim of this study was to analyze the ganglionic effect
of P4, through SON, on the luteal expression of 3β-HSD, P4 synthesis enzyme,
Bax and Bcl-2, regulator factors of the
way intrinsic of apoptosis in pregnant rats
on 21 day using the ex vivo CG-SON-ovary system.
The system was incubated in Krebs
Ringer at 37ºC,
keeping CG and ovary connected by the SON, in separate compartments. P4 (10-5M) was added in
the ganglion compartment [(P4)g], At the end of incubation (180 min), the luteal expression of 3β-HSD
was studied by Western Blot and the expression of Bax and Bcl-2
was analysed by RT-PCR. Student´s test with a statistical significance of p<0.05 was used.
[(P4)g] did not modify
significantly the expression of the parameters of luteal regression studied in
this work. In conclusion, the luteotrophic effect of
P4 through the peripheral neural
pathway would be mediated by inhibition of 20α-HSD enzyme and by modulation of
extrinsic pathway of apoptosis.
