Resumen:
It is of great concern the presenceof contaminants in commercial products, in particular in pharmaceutical drugs.Knowledge of the stereochemistry of these contaminants is of the utmostimportance due to the different toxicity presented by their stereoisomers.Specifically, β-blockers are sold and administered asa racemic mixture and although the (S) enantiomer is the pharmacologicallyactive, the (R) enantiomer is the one which has toxic effects [1].Electrokineticchromatography (EKC) has become a widely accepted technique forelectrophoresis-driven separations.Pseudostationary phases (PSPs) have the advantage ofintroducing a novel interaction phase for the analytes, which greatlysimplifies column exchange and circumvents contamination inherent with complexmixtures. In this work we implement nanodiamonds (NDs), a class of carbonnanomaterials with most particles lying between 2 and 20 nm in diameter, aspart of PSP.Theirpotential is based on its excellent mechanical and optical properties, highsurface areas and tunable surface structures. They are also non-toxic, whichmakes them well suited to biomedical applications [2].In this work we proposed a methodology to separate aracemic mixture of emerging pollutants, so we focused the first analysis in oneof β-blockers most commonly used today worldwide propranolol. We performed a nano-EKC methodology adding NDs-albumin, a proteinwell known as chiral selector, to the background electrolyte (BGE) to generatea PSP system. Beckman P/ACE MDQ instrument was used. The dimensions of the capillary used was 75 µm i.d. and15 cm total lenght capillary and pseudostationary fase system consisted in with40 mM sodiumtetraborate(pH=7,9) and 5% NDs-albumin bonded by electrostaticinteraction between carboxylated nanodiamonds groups and amine proteins groups. The proposed method wassuccessfully employed for the separation of the R-propanolol and S-propanololenantiomers. The use of NDs-albumin as PSP in nano-EKC improves the resolutionsubstantially with small amounts of reactive.References:1. Scriba GKE, J. Pharm. Biomed. Anal.,2011, 55, 688?701. 2.
