Melanoma is a highly aggressive type of cancer, characterized by malignant transformation of melanocytes and a high relapse rate, multiple drug resistance and, low survival rate. This study was focused on the development of composite chitosan matrices functionalized with different proportions of Aloe vera with potential application in biomedicine. In this sense, the ability to inhibit the viability of malignant melanocytic cells (SK-MEL 28) and keratinocytes (HaCat cell line) in culture was evaluated.To obtain a composite filmogenic solution, different proportions of Aloe vera gel were combined with chitosan solution. The biomaterials presented hydrophilic properties, with an average contact angle of 84.3º and 43º for chitosan and composite film 75/25 (A/CH), respectively. Spectroscopic studies by ATR-FTIR confirmed that the presence of C=O stretching vibrations of carbonyl groups in the region of 1740 cm-1 is possibly due to the functional groups of Aloe vera reacted with chitosan, showing a molecular bond. XRD pattern confirmed a weaker and broader peak, indicating that an amorphous structure prevailed after conjugation with Aloe vera, .Functionalized CH films with different proportions of Aloe vera inhibited the viability of SK-MEL 28 cells in 90% after 24 h of incubation. The 0.1 mg/mL and 1 mg/mL Aloe vera extracts were less effective, inhibiting the cell viability by 12 and 25%, respectively. The functionalization of chitosan-based films with Aloe vera enhanced the anticancer activity of Aloe vera. Based on the results obtained, the films have suitable properties for topic melanoma applications.
