Most of the neurodegenerative pathologies that affect the posterior segment of the eye are characterized by being chronic, devastating and responsible for the majority of visual dysfunctions worldwide. The design of controlled drug release systems to avoid frequent invasive interventions is an interesting alternative, being albumin nanoparticles (Np-ASH) a potential strategy because they are non-toxic, biodegradable and without antigenic activity. On the other hand, there is evidence that suggests that Melatonin has antiapoptotic and antioxidant properties, thus protecting cells from damage caused by free radicals. The objective of our study was to design, formulate and characterize NP-ASH to vehicle Melatonin.The obtaining of NP-ASH-Mel was carried out through different methods to which a subsequent physicochemical characterization was carried out. A combined method between desolvation and thermal stabilization processes was chosen as the most auspicious as it is easily scalable and dispenses with the use of crosslinking agents. Whitish colloidal dispersions with a pH close to neutrality, nano-sized, monodisperse and highly stable population were obtained. In addition, yield values of the process greater than 80% were obtained, while in-vitro release tests demonstrated sustained release of the drug for more than 24h. These auspicious results will allow us to advance in ex-vivo and in-vivo trials and evaluate the potential of our system in the treatment of ocular neurodegenerative pathologies.
