Postprandial hyperglycemia is a risk factor for diabetic complications. a-glucosidase inhibitors delay the absorption of ingested carbohydrates, reducing the postprandial glucose and insulin peaks. In previous studies, we have demonstrated that Smallanthus macroscyphus is a plant species that has promising properties for the treatment of diabetes. This work was undertaken to evaluate their possible anti-hyperglycemic mechanism. In vitro assays showed that S. macroscyphus leaves decoction (DEC) and its active compound, Polimatin A (PA) significantly inhibited the activity of a-glucosidase enzyme in a dose-dependent manner. In vivo experiments in normal and diabetic rats, oral administration of DEC and PA caused a rapid decrease of hyperglycemic peak after oral glucose load. Oral administration of DEC and PA, prior to sucrose overload, caused a significant decrease in blood glucose, demonstrating the inhibitory effect of the a-glucosidase enzyme in vivo. The treated rats with DEC showed better response anti-hyperglycemic which the pure compound. This study demonstrates that decoction of S. macroscyphus leaves and PA exerts the anti-hyperglycemic effect through the mechanism of inhibition of a-glucosidase enzyme in the intestine. However, other targets of action are not excluded.
