1. Abstract MicroRNAs (miRNAs) are key post-transcriptional regulators of gene expression in many cellular pathways, and play critical functions in developmental and pathological processes. Thus, alterations in the levels of individual or entire families of miRNAs are associated with the pathogenesis of a wide range of human and animal diseases. Indeed, disease-associated miRNAs represent a new class of targets for the development of miRNA-based therapeutic modalities. The interest in miRNA research has accelerated the development of several in vitro and in vivo experimental approaches for probing miRNA functions by means of miRNA interference. This chapter will summarize and assess the technologies that have demonstrated their efficacy and reliability in inhibiting and enhancing miRNA expression. Furthermore, we describe the most commonly used in vivo delivery strategies for miRNA interference. Finally, we critically summarize recent progress of therapeutics based on miRNA interference and their impact in human diseases.
