The optimization of biological processes requires knowledge of the physicochemical and microbiological characteristics of the inoculum used and the substrate to be treated. The objective of the present study was the characterization of activated sludge and vinasse for starting a UASB reactor. The selection of the activated sludge was performed taking into account the amount of total suspended solids and specific methanogenic activity. Five sludges from different sources were analyzed. The sludge from storage lagoons vinasse presented the best conditions to inoculate the reactor. Vinasse was selected as the only carbon source. It was characterized by determining COD, pH, temperature, conductivity and total phosphorus. The necessary amount of phosphorus was added to achieve the ratio of COD: N: P of 500:7:1 and promote granulation process. The selection of an inoculum from the vinasse treatment, shorten the time of adaptation of microorganisms. Tests on samples obtained from methanogenic activity sectors allowed us to select sludge to be used subsequently into a UASB reactor. Different vinasse samples were characterized. They will be used in the granulation process of the reactor, for subsequent study of the same starting and stabilization. To achieve use the stillage in UASB reactor types should make some modifications: pH and phosphorus added for effective treatment in these reactors
