Thermodynamically controlled covalent chemistry opens new possibilities for the synthesis of macrocycles. The use of template molecules can alter the outcome of a reaction in a significant way, favoring the concentration of previously unexpected products. Examples where covalent reactions under thermodynamic conditions are combined with organic or inorganic templates and used for the high yield preparation of O-, N-, and O,N-macroheterocycles will be discussed.
