Resumen:
SHR with cardiac hypertrophy (CH) were treated with the NHE-1 inhibitors cariporide or BIIB723. NHE-1 blockade induced CH regression evidenced by a decrease in left ventricular (LV) weight/body weigh (LVW/BW) and myocardial BNP and calcineurin (Cn) Ab expression, without changing blood pressure. The reduction in LV mass without changes LV pressure significantly increased peak-systolic stress in treated SHR. Despite this, fractional shortening did not decrease and even an increase in mid-wall shortening was detected, suggesting that a positive inotropic effect develops after CH regression. This was not due to an intrinsic effect of the NHE-1 inhibitors. NHE-1 inhibition-induced CH regression was accompanied by decreased CnAb expression and maintained cardiac function despite the increase in LV wall tension, suggesting a cause-and-effect relationship between Cn expression and contractility.
