We introduce a general framework for biological systems that describe Modifications of type Enzyme-Substrate or Swap with Intermediates, which we call MESSI networks. Many post-translational modification networks are MESSI networks. For example: the motifs in [5], sequential distributive multisite phosphorylation networks[17], sequential processive multisite phosphorylation networks [1], or phosphorylation cascades or the bacterialEnvZ/OmpR network in [22]. We prove that, under mass-action kinetics, MESSI systems are conservative. We simplify the study of steady states of these systems by explicit elimination of intermediate complexes (inspired by[4, 6, 24]) and we describe an important subclass of MESSI systems with toric steady states [17].
