Preeclampsia (PE) is associated with deficient trophoblast invasion of spiral arteries. The direct cause remains unknown. Altered placental villous angiogenesis and poorly developed fetoplacental vasculature can affect the transport functions of the syncytiotrophoblast (hST). Previously, we reported an increase of Aquaporin 9 (AQP9) expression in hST of preeclamptic placentas. AQP9 is involved in water and neutral solutes fluxes. It is known that insulin downregulate AQP9 because its promoter contains the negative insulin response element. Here, we investigate whether Human chorionic gonadotropin (hCG), Insulin-like growth factor 1 (IGF-1) and insulin may modulate AQP9 expression. Insulin, IGF-1 and hCG levels were measured in serum of preeclamptic (n=13) and health gestants (n=12). Explants from normal and preeclamptic placentas were cultured at different concentration of insulin and hCG. Western blot assays and immunohistochemistry were used to quantify AQP9 expression. In PE hCG and Insulin levels were significantly higher (p
