Amniotic fluid is essential for normal fetal growth, movement and development. The fetal membranes play important roles in amniotic fluid homeostasis. The expression of aquaporins (AQP) 1, 3, 8 and 9 has been described in fetal membranes. Previously we have demonstrated the functionality of these AQPs suggesting that they may mediate amniotic water fluid regulation. Here, we studied if AQP9 expression is regulated by progesterone (P4) human chorion. Methods: Human chorion explants were cultured with 300 ng/mL progesterone (P4). They were mounted as a diaphragm between two lucite chambers and the net transepithelial water movement (Jw) was recorded minute by minute. Semi-quantitative RT-PCR was performed to quantify AQP9 mRNA and inmunohistochemistry was made to determine its localization.
