Resumen:
Objectives: Previously, we found that AQP3 shows a different pattern expression in amnion cells exposed to osmotic stress, suggesting that the mechanism of regulation of this protein is different from other AQPs. Thus, our purpose was to study the regulation of AQP3 in amnion cells.Methods: Amnion-derived WISH cells were cultured in hypo (150 mOsm) and hyperosmolar (400 mOsm) conditions. AQP3 expression was examined. Levels of phosphorylated ERK (pERK), JUNK (pJNK), and p38 (p-p38) were studied. Nf-ĸB and tonEBP expressions were assessed in nuclear and cytosolic fractions. AQP3 levels were analyzed after the inhibition of Nf-ĸB and tonEBP pathways with Sodium Salicylate and Cyclosporine-A, respectively. Results: In amnion cells cultured in hyperosmolarity AQP3 levels increased. On the other hand, when cells were cultured in hypoosmolarity, AQP3 decreased.
