A full-factorial design was used to assess the influence of ion strength and Triton X-114 concentration for the extractive purification of a lipase frorn Acinetobacter calcoaceticus AAC 323-l in temperature-induced aqueous two-phase systems. The concentration for optimal extraction was found at 0.75 M NaCl and 3% Triton X-114 concentrations, as shown by surface response analysis. Based on these results, a rapid lipase purification procedure from culture supernatant was developed. Such procedure allows direct extraction from culture broths in three successive steps with no additional operations. This technique enabled us to extract 68% of the enzyme and achieved a purification factor of 41.
