The chromosome damage was analyzed in the progeny of mammalian cells exposed to the radiomimetic compound, bleomycin (BLM) by using the Fluorescence in situ hybridization (FISH) technique with a PNA telomeric probe. The aim of this investigation was to evaluate if BLM induces delayed chromosomal instability at telomeres. FISH with the PNA (TTAGGG)n probe was applied on chromosome preparations of rat cells derived from adipose tissue (ADIPO-P2 cell line). Cells were exposed in the log phase of growth to 2.5 µg/ml of BLM during 30 minutes at 37°C. Cytogenetic analysis revealed a significant increase in the frequency of chromosomal aberrations at 18 h and 10 days after treatment in ADIPO-P2 cells treated with BLM compared with unexposed cells, this finding showing that BLM has a delayed clastogenic effect, which lasts for at least 10 days after treatment. Our results also indicated that BLM induced delayed telomeric instability in ADIPO-P2 cells, revealed by the presence of incomplete chromosomes and additional telomeric FISH signals in the chromosomes of the exposed cells.
