Possitive effect of 17b estradiol on movement disorders associated with Parkinson Disease

Software development and implementation for motor parameters? evaluation. Parkinson Disease (PD) generates a profound deficit in dopaminergic neurons of the nigrostriatal pathway with pathological movements such as tremors, stiffness, bradykinesia and gait disorders. Because PD affects men more than women, it is believed that female sexual steroids could act as neuroprotectors against this disease. We use an experimental male rat model of hemiparkinsonism induced by 6-OHDA to investigate 17β-Estradiol (E) effect on movement disorders on different stages of the neurodegenerative process associated with PD. We implement bioinformatic softwares to automatically assess motor behavior; open field test (OFT) to evaluate locomotor performance, forced swimming test (FST) to measure movement modifications and stepping test (ST) to calculate forelimb akinesia parameters. MATERIALS AND METHODS: On postnatal day 60 (PND), adult male Sprague Dawley rats were anesthetized and received 6-OHDA= 2 μg/μl or vehicle (V) in left corpus striatum. From PND 67 to 77, they received chronic treatment with E= 0.1 μg/kg/day s.c, or corn oil (O) as vehicle. Groups: HPE (6-OHDA, E, n=10); HP (6-OHDA, O, n=10); C (V, O, n=10); E (V, E, n=10). OFT was conducted on PND 74 without drug induction (NDI) and, on PND 88 after amphetamine injection (AMPH= 1mg/kg i.p). After 48h FST NDI was evaluated. ST  NDI was performed on PND 116.
