Background and Objectives: In recent years, the finding of a potentially lethal side effect, known as Torsades de Pointes, in a wide range of medications led to the development of viability benchmarks that assists in the decision making on drug cardiotoxicity. Unfortunately, current technics are not enough efficient and many safe drugs are withdrawn while many proarrhythmic drugs are realeased. Methods: Herein,abnormal ventricular heterogeneity is induced by d-Sotalol supply in eleven In-vitro rabbit hearts immersed in a multi-electrode chamber. We propose novel velocity cardiomarkers obtained by quaternion methods, which have been shown to be highly efficient in coronarydiseases. Through statistical and classification methods, we find the more significant dynamic features.Herein, a Sotalol challenge with 34 patients with and withouthistory of torsades de pointes is used to evaluate differential dynamic features obtained by a quaternions method. We propose several dispersion cardiomarkers using a spatial approach in order to detect abnormal drug-induced changes of heterogeneity. Results: Novelcardiomarkers not only achieves a higher significance level than current indices, about two order of magnitude, but also they can detect sotalol-induced changes that onlyaffect at-risk patients. Conclusions: This novel technique of ECG processing complement current studies of drug cardiotoxicity. Given the high significance levels of ourcardiomarkers, a profundization of this research line will undoubtedly be able to contribute to the development of safe medication therapies.
