p { margin-bottom: 0.25cm; line-height: 120%; }a:link { }Bacteriocins arebacterial antimicrobial peptides of ribosomal synthesis active onphylogenetically related organisms.In previous studies we produced anew hybrid peptide named PedA1-GGG-ColV whit broader antimicrobialspectrum.The aim of this work was to evaluate antimicrobial activityagainst different pathogenic strains and determinate the sensitivityof peptide to proteolytic enzymes present in the stomach andintestinal contents.To evaluate the antimicrobial activity, we spreada standardized amount of bacteria uniformly on the surface of a LBagar plate to obtain a bacterial lawn.Then 10 ml of a solution ofeach bacteriocin were spotted on plates.To determine the sensitivityof the hybrid bacteriocin to proteolytic enzymes,10 μl of peptidewere spotted onto LB plates and 10 μl of proteolytic enzymessolution was placed 1 cm away from the cultured medium drops.Theplate was covered with 4 ml of 0.6% agar previously inoculated withthe indicator strain. After overnight incubation at 37°C, the growthinhibition zones around the drops of hybrid media were recorded. Weinterpreted the shapes of growth inhibition zones around these hybridas suggesting that digestive enzymes diffuse and decreasing theefficiency of indicator strain growth inhibition.PedA1-GGG-ColVdisplayed an antibiotic effect on most of the tested strains andresult inactivated by digestive enzymes in vitro.
