Functional activity of spermatozoa from mice treated with nonsteroidal-antiinflammatory drugs.

  Introduction: Several studies indicate that drugs interfering with prostaglandins metabolism, such as nonsteroidal anti-inflammatory agents, may affect male reproductive function. Nevertheless controversial resulta have been reported about this matter. In the present work we studied the influenec of ibuprofen (IBU) or piroxicam (PIR) on motility, reponse to hypoosmotic shock, acrosome integrity and in vitro fertilization rate (IVF) of epididymal mouse spermatozoa. The rate of pregnancy after mating was also quantified. Material and methods: IBU (0.56 mg/100g/day) or PIR(0.0287mg/100g/day) were injected (ip) to sexually mature male mice during 60 days. After this period, the animals were sacrified and epididymal spermatozoa were suspended in Tyrode´s médium. Control animals (C) received daily injections of the respective vehicles (propylene glycol and dimethyl sulfoxide). In order to investigate the in vitro effects of 10-7, 10-5 or 10-3 M IBU or PIR, the drugs were added after 60 min incubation and the functional parameters were measured 20 min thereafter. Results: In vivo administration of PIR only modified IVF 72.4% in C vs 88.7%, p<0.05) and the rate of pregnancy after mating (78% in C vs 43%, p< 0.05). Preliminary results indicate similar tendency in the experiments performed with IBU. In vitro addition of 10-3M IBU significantly diminished the percentage of acrosome intact gametes from 59.0 ± 7.1% after 60 min incubation to 38.6 ± 3.5%, n08, P< 0.05. When homologous oocytes were inseminated with spermatozoa pretreated with 10-3M IBU, the IVF dropped from 64% to 52%. Similar results were obtained with 10-3M PIR. Conclusions: The ressults suggest that PIR or IBU modify the function of male gametes. These influences vary when in vivo or in vitro administration is considered. So, afetr 60 days daily injections, the in vitro fertilization capacity seems to be enhanced, while the in vivo fertilization rate decreases. The blockade of the endogenous prostaglandins síntesis may explain these observations. The reduced acrosome integrity after 20 min in vitro treatment with 10-3 M PIR or IBU could be responsible for the reduced IVF.
