Resumen:
Venlafaxine hydrochloride(VEN)isanantidepressantdrugwidelyusedforthetreatmentofdepression. The purposeofthisstudywastocarryoutthepreparationandsolidstatecharacterizationofthepure polymorphs (Forms1and2)andthepolymorphicidentification andquantification offourcommercially- availableVENrawmaterials.Thesetwopolymorphicformswereobtainedfromdifferentcrystallization methods andcharacterizedbyX-rayPowderDiffraction(XRPD),DiffuseReflectance InfraredFourier Transform(DRIFT),RamanSpectroscopy(RS),liquidandsolidstateNuclearMagneticResonance(NMR and ssNMR)spectroscopies,DifferentialScanningCalorimetry(DSC),andScanningElectronMicroscopy (SEM) techniques.ThemaindifferenceswereobservedbyDSCandXRPDandthelatterwaschosenas the standardtechniquefortheidentification andquantification studiesincombinationwiththeRietveld method forthecommercialrawmaterials(VEN1?VEN4) acquiredfromdifferentmanufacturers. AdditionallyForm1andForm2canbeclearlydistinguishedfromtheir 13C ssNMRspectra.Through the analysis,itwaspossibletoconcludethatVEN1andVEN2werecomposedonlyofForm1,whileVEN3 and VEN4wereamixtureofForms1and2.Additionally,theRietveldrefinement wassuccessfully applied toquantifythepolymorphicratioforVEN3andVEN4.
