Resumen:
Phalloidin has been used extensively to study the distribution of F-actin in cells and tissues. Through the use of fluorescence photo-oxidation we have employed phalloidin conjugates to examine the precise cellular and subcellular localization of F-actin in the central nervous system. Correlative fluorescence light microscopy and transmission electron microscopy studies of F-actin distribution are made relatively easy using this method. Because phalloidin-eosin is smaller than other markers, this method allowed us to analyze the three-dimensional localization of F-actin using high-resolution light microscopy and electron tomography. Thus this technique is an excellent tool to study F-actin rich structures across a single,identified cell.
