Resumen:
gd T cells act as sensors of cellular stress and infection. They recognize danger and pathogen-associated molecular patters, such asphosphoantigens. Uric acid is a cellular stress signal, which is released from dying cells and promotes the generation of innate andadaptive immune responses. The inflammatory effect of uric aciddepends on its precipitation into monosodium urate (MSU) crystals. MSU crystals activate different intracellular pathways, i.e. p38MAPK, Syk kinase and PI3K. We have previously reported that MSUcrystals activate gd T cells and that this activation was reduced inpresence of a Syk tyrosine kinase inhibitor. Now, we aim to investigate more in detail the participation of Syk tyrosine kinase during theintracellular signaling triggered by MSU crystals in gd T cells. To thispurpose, gd T cells were purifed from human peripheral blood mononuclear cells by using an anti-TCR gd MicroBead isolation kit. Afterpurifcation, gd T cells were incubated or not with MSU crystals (200µg/ml) at different time points, and then we analyzed the expressionof phospho(p)-Syk (the active form of the tyrosine kinase) by immunofluorescence and confocal microscopy analysis.
