Resumen:
Trophoblast cells (Tb) interact with different maternal immune cell populations at early pregnancy promoting a local anti-inflammatory and tolerogenic response. Neutrophils (neu) are highly detected in human uterine tissues at the secretory phase and decrease in the first trimester of pregnancy. Vasoactive intestinal peptide (VIP) is a pleiotropic peptide with immunomodulatory effects through its action on VPAC1 and VPAC2 receptors. Our aim is to evaluate the effect of VIP and Tb derived factors on neu activation. Whole blood was obtained from healthy donors and neu purified on Ficoll-Triyosom gradient and dextran sedimentation. VPAC2 receptors are expressed in neu and a trend increase of VPAC2 expression was observed after 4h incubation with conditioned media (CM) from human first trimester Tb (Swan-71 cell line). The effect was higher when neu were incubated with CM obtained from Tb treated for 18h with 10 nM VIP (CM*VIP). VIP, CM and CM*VIP decreased neu activation induced by fMLP or PMA resulting in lower CD11b expression and lower release of reactive oxygen species determined by FACS.
