Exploring binding interactions in living cells through advanced fluorescence microscopy and numerical simulations.

The glucocorticoid receptor (GR) is a ligand-activated transcription factor. Inthe nucleus, freely diffusing GR partitions into subpopulations interactingwith different subcompartments, such as discrete foci and DNA targets withvariable specificity. This partition can be tuned by the use of different GRligands and a GR coregulator like NCoA-2, and could ultimately affect GRtranscriptional activity.We have previously studied these dynamic interactions by advanced fluorescence methods in the nucleus of living cells. However, the interpretation of fluorescence data is not always simple, and sometimes different techniques lead to different results.In this sense, simulations can help to interpret fluorescence data. We used the Fluorescence Emission Recipes and Numerical routines Toolkit (FERNET) to model complex biological systems and simulate the fluorescence data acquisition. Then, we analyzed the data as it came from experimental measurements and we contrasted the results with the expected ones.
