Assessment of biodiversity in Chilean cattle using the distribution of major histocompatibility complex class II BoLA-DRB3 allele

Bovine leukocyte antigens (BoLA) are extensively used as markers for bovine disease and immunological traits. In this study, we performed genotyping about BoLA-DRB3 allele for 888 of various cattle breed such as 99 Red Angus, 100 Black Angus cattle, 81 Chilean Wagyu cattle, 49 Hereford cattle, 95 Hereford x Angus, 71 Hereford x Jersey, 20 Hereford x Overo Colorado, 113 Holstein, 136 Overo Colorado, and 124 Overo Negro cattle, and determined BoLA-DRB3 allele frequencies. 46 BoLA-DRB3 alleles were identified in 10 Chilean breeds and each breed had 12 to 29 kinds of BoLA-DRB3 alleles. The highest number of alleles (29 alleles) was detected from Overo Negro, which is considered in Chile as ?Old type? European Holstein Friesian descendants. On the other hand, only 21 alleles were detected from Holstein, which is considered as ?Present type? Holstein Friesian cattle. Chilean cattle breed were compared by phylogenetic and principal component analysis (PCA) using four Japanese breed as out group. Structure of phylogenetic tree showed that Red Angus and Black Angus positioned at same clade, mix breed were positioned near from original breed, and Holstein from Chile was positioned in same clade of Holstein in Japan. Therefore it can be thought that this tree was well explanation of breed constructed history. This tree also showed that Overo Negro placed near from within Holstein, followed the historical data that Overo Negro would be the ?Old type? Holstein Friesian cattle. PCA analysis showed that the breed originated in British Island (Angus, Hereford, and Jersey) was divided from breed originated in European continent (Holstein, Overo Colorado and Ovelo Negro) at the point view of PC1 value, which mainly computed from the frequencies of 16 BoLA-DRB3 alleles. These allelic information of MHC gene was important for investigating the relationship between host factors and disease association.
