Induction of apoptosis by hyaluronan oligosaccharides is independent on the transcription factor NF-kB

Abstract Several studies indicate that hyaluronan (HA) oligosaccharides can modulate the growth and the cell survival in solid tumors, however, no studies have been undertaken to analyze the effect of HA oligomers on T lymphoma cells. We have previously shown that native HA enhances basal NFkB activity found in lymphoma cell lines. Since NFkB is the major transcriptional factor involved in cell survival and anti-apoptotic pathways in lymphoma cells, we hypothesized that HA may be responsible for such increase in cell survival. We showed that hyaluronan oligomers were able to induce apoptosis in these cell lines, although they failed to inhibit constitutive NFkB activity and modulate IkBa protein levels. However, no effect of native HA on T cell survival was found. Besides, we used BAY 11-7082 as control of NFkB inhibition, which was able to induced apoptosis. Our results suggest that hyaluronan oligosaccharides could induce apoptosis independently of NFkB signaling pathway.
