Sulfated N-linked-oligosaccharides have been reported as components of glycoproteins from mammalian cells and a few other organisms: birds, Dyctiostelium discoideum and some viruses. In most cases, these glycans are involved in adhesion processes. In the present work we report for the first time the presence of sulfated N-linked oligosaccharides as components of a lysosomal glycoprotein of the parasitic protozoan Trypanosoma cruzi. Analysis by HPAEC-PAD, of the oligosaccharides, released from cruzipain by PNGase F digestion, before and after mild acid hydrolysis, suggested the presence of an acidic group different from sialic acid previously reported. Phosphate groups had been discarded after in vivo labelling with 32Pi. Thus, the presence of sulfate groups was investigated by different methods. The UV-MALDI TOF analysis of the released oligosaccharides, in the negative-ion mode, using nor-harmane as matrix, showed the presence of sulfated high-mannose type oligosaccharides.
