Cocaine, caffeine and their combination alters the pacemaker activity of somatosensory thalamic neurons

Caffeine is a common stimulant drug worldwide consumed that can be found in a range of commercially available products. Cocaine is an addictive stimulant drug highly dangerous that can cause dependence, addiction and long-term consequences when abused. In South America there is an endemic use of a drug known as ?PACO? in which it was found quantities of caffeine mixed with cocaine. Caffeine is the most widely consumed psychoactive substance and has complex pharmacological actions in brain, the most prominent is that it reversibly blocks the action of adenosine on its receptor and consequently prevents the onset of drowsiness induced by adenosine, it can elicits physiological responses in a variety of cell types by triggering the mobilization of Ca 2+ from intracellular organelles via ryanodine receptors. In the last years the action of caffeine in other targets of the brain has been studied. Kramer & cols. (1994) found that caffeine inhibits the L-type Ca2+ channels. We studied the effects of an acute binge administration of caffeine, cocaine of an acute binge administration of caffeine, cocaine or both combined on synaptic properties of Ventrobasal neurons(VB). Male C57/BL6 mice were treated with cocaine (10 mg/kg; 3 inj. 1 hr apart, i.p.), caffeine (5mg/kg; 3 inj. 1 hr apart, i.p.), their combination (CC) (10 mg/kg Coc + 5 mg/kg Caf 3 inj. 1 hr apart, i.p.), vehicle (Veh: Saline solution).). We recorded VB neurons 24 hs after last binge injection using whole cell patch-clamp and investigated the effect of these treatments on glutamatergic evoked neurotransmission and action potential?s parameters . We also evaluated the total VB calcium concentration in VB neurons with fura-2 ratiometric calcium imaging registration before and after bath application ofcaffeine (20 mM) and analyze the effects of bath applied Caffeine (20mM).Kramer et al.
