Resumen:
The aim of this work was the design of phytomedicine tablets using as active component a Cáscara sagrada (Rhamnus purshiana) spray dry extract (SDE) with good flow properties for direct compression (DC). The extract was spray drying in a Büchi B-290 Mini Spray. The drying conditions were: drying air inlet temperature 130º C, atomization air volumetric flowrate 400 l/h, feed volumetric flowrate 15%, drying air volumetric flowrate 80% and dispersion concentration 7.32 % (w/w), with 1:1 colloidal silicon dioxide (as drying aid):solid residue ratio. The actives components (hydroxyanthracene derivatives and cascarosides) quantitative determination was assay. The four solid pharmaceutical formulations containing the SDE and common excipients for DC were formulated used a 22 factorial experimental design. The physical-mechanical properties (repose angle and Carr´s Index), the hardness, friability, disintegration time and dissolution test were evaluated. The four formulation presented good flow properties and acceptable hardness, friability and disintegration time. The dissolution test showed, in some formulations, a rapid release of the actives components (approximately 85%, before five minutes). The release was greatly influence by the present of the different excipients. The following formulation: SDE (146 mg), Lactosa CD (70 mg), Avicel PH101 (70 mg), Acdisol (70 mg) and magnesium stearate (4 mg) showed the best pharmaceutical performance.
