Cytotoxic effects of merocyanine 540 (MC) and photoactivated pMC540 in human carcinoma cell lines

Resumen:
Photodynamic theray (PDT), an alternative treatment for cancer, involves a photochemical reaction between a sensitizar, light and axygen to generate cytotoxic reactive oxygen species (ROS) leading to cell death. Another strategy implies the pre-activation of the sensitizer prior to its use where photooxidation or breakdown into smaller substances biologically active could result by light irradiation. The presens study was carried out to characteriza pre-activated merocyanine 540 (pMC540) products by UV-Vis, fluorescence and FT-IR spectroscopy and compare their prperties and cytotoxic in human carcinoma cell lines respects to MC540. Cytotoxic was determined by crystal violet assay. Our results suggest that photoactivation breaks down MC540 generating al least two pMC540 products which cytotoxicity is dependet on the cell line type. Either MCF-7 or Caco-2 cells were not susceptible to MC540, pMC540 showed to be dose-dependet for MCF-7 cells. Caco-2 colorectal adenocarcinoma cells were not susceptible to pMC540 photoproducts. We hypothesize that these differeces could be derived from the differential interaction of MC540 and/or pMC540 on different cell membranes constituents. Further assays and analysis are ongoing to strengthen our hypothesis.
