Evaluation of anticancer activity of selected polypore medicinal mushrooms: Grifola gargal (GG), G. sordulenta (GS), G. frondosa (GF) and Ganoderma lucidum (GL) in two kind of biomass sources: fruiting bodies (FB) and biotransformed wheat grains (BWG)

Background:Grifola gargal (GG), G. sordulenta (GS), G. frondosa (GF) and Ganoderma lucidum (GL) are polypore mushrooms regarded as a valuable source of food and medicine in Asian countries like China and Japan, in central and eastern Europe, and by the inhabitants of the patagonian natural forests of Argentina and Chile. Recent scientific research supports their use as functional foods with antioxidant, antigenotoxic and anticarcinogenic activity. , , Methods:Two kind of biomass sources were employed: fruiting bodies (FB) and mycelium biotransformed wheat grains (BWG). Ethanolic extracts were prepared by Soxhlet method from 12 milled samples (1 commercially available GL FB, 2 GF BWG, 3 GL BWG, 4 wheat grains (control), 5 GS BWF, 6 GG BWF, 7 GG FB, 8-12 GL FB cultivated on sunflower or with rice side streams with different supplements) produced by Cerzos-UNS/Conicet (Argentina). Anticancer activity was evaluated by means of in vitro MTT assay in human colon carcinoma HT-29 cells and a normal colon epithelial cell line. Results:The most effective extracts were obtained from Ganoderma lucidum E47 (samples 8-12). These extracts showed a prominent antiproliferative activity against colon carcinoma HT-29 cells and lack of cytotoxicity in normal colon epithelial cell culture. It should also be emphasized that extracts obtained from G. lucidum cultivated on sunflower seed hulls (samples 8-12) had a higher anticancer activity than the commercially available fungi (sample 1).Conclusion:Based on these results, samples 8 and 10 are the most promising for future research in the context of the high antiproliferative activity against cancer cells and the low cytotoxicity against normal cells.
