Resumen:
Most of the neurodegenerative pathologies that affect the posterior segment of the eye arecharacterized by being chronic, devastating and responsible for the majority of visualdysfunctions worldwide. There is evidence that drugs with antiapoptotic and antioxidantproperties are capable of protecting cells from damage caused by free radicals. The chosenantioxidant has poor solubility and low permeability, so the design of protein nanoparticles(Np-Pro) is an interesting alternative, a potential strategy because they are non-toxic,biodegradable and without antigenic activity. The objective of our study was to design, formulate and characterize NP-Pro to vehicle an antioxidant agent (Aox). The obtaining of NP-Pro-Aox was carried out by different methods to which a subsequentphysicochemical characterization has been made. A combined method between desolvationand thermal stabilization processes was chosen as the most auspicious, since it is easilyscalable and dispenses with the use of crosslinking agents. Whitish colloidal dispersions with apH close to neutrality, nano size, monodisperse and highly stable population were obtained. Inaddition, process yield values greater than 80% were obtained, while in vitro release testsdemonstrated a sustained release of the drug for more than 24 h. These auspicious results willallow us to advance in ex vivo and in vivo trials and evaluate the potential of our system in thetreatment of ocular neurodegenerative pathologies.
