Toxicity of cypermethrin on retinal and gill cells of zebrafish (Danio rerio): apoptosis, genotoxicity and alteration in the activity and gene expression of antioxidant enzymes

Resumen:
Cypermethrin (Cyp) is a broad spectrum insecticide, widely used both in domesticagricultural applications, combining its excellent ability to control these organisms postulatedwith low toxicity to mammals and other vertebrates. In this work, we analyze the effect ofCyp on the retinal and gill cells adult zebrafish. The animals were exposed to 0,3 μg/L and0,6 μg/L and the vehicle of dissolution pyrethroid (control) for 3, 6, 9 and 12 days. Theevaluation is through by means of histological techniques; comet assay; and changes inboth the enzymatic activity and gene expression of enzymes CAT and SOD. Nomorphological changes were observed in the retinal layers with 0,3 μg/L, while 0,6 μg/Lcaused the appearance of apoptotic figures in the photoreceptor layer. The cells of the retinaand gill exposed to both concentrations exhibit significant increases in the rates of DNAdamage after nine days exposure compared to respective controls. The enzymatic activityalso verified significant increases after nine days with both concentrations. The analysis ofgene expression of both enzymes produced by sqPCR a pattern according to the activity ofenzymes studied compared to the β-actin gene used as housekeeping. The changes foundwere related to the imbalance of reactive oxygen species, resulting in generation of oxidativestress in both cell types. This information allows further study of the possible mechanismsof action of Cyp in vertebrates.
