Excitatory (i.e.
glutamatergic) neurotransmission in nucleus accumbens (NAcc) is relevant for
cocaine-induced long-term neuroadaptations. The involvement of one type of
glutamate receptor, the AMPA receptor (AMPAR), in cocaine-induced changes in
the NAcc has been shown by our lab and by others. Narp (Neuronal activity
regulated Pentraxins) is an extracellular matrix protein that plays a role in
aggregating AMPAR at pre- and post-synaptic level. This aggregation, also
called clustering of the AMPAR, is believed to facilitate the response of the
receptors. The present study was designed to determine whether Narp-induced
AMPAR clustering would participate in cocaine-induced behavioral
neuroadaptations. We compare cocaine effects in mice with Narp (e.g. Narp wild
type mice) and without Narp (e.g. Narp knock out mice). Among the cocaine
effects we measured were cocaine rewarding properties by conditioning place
preference and stimulant properties by a locomotor sensitization paradigm. Narp
knock out mice displayed higher cocaine place conditioning and locomotor
sensitization compared to Narp wild type mice. The behavioral results suggest
that Narp-induced AMPAR clustering could have a compensatory role in cocaine
long-term neuroadaptations. The molecular mechanisms underlying these possible
compensatory effects are under investigation.
