The Arabidopsis thaliana cysteine desulfurase AtNFS1 has an essential role in cellularFe-S cluster assembly. In recent studies, we reported a relationship between thisbiosynthetic pathway and iron metabolism. We also reported that FH, a proteinfunctionally associated with NFS1, is essential for heme biosynthesis. To furtherunderstand the relationship between the Fe-S pathway enzymes and the hemebiosynthesis, we studied the heme content, the expression levels of the heme pathwayenzymes and the catalase activity of two mutant lines (KD501 and KD681) and twooverexpressing lines (MycA and MycB) of AtNFS1 in Arabidopsis thaliana. WhileMycA and MycB showed an increase of 30% of heme content, an increase of catalaseactivity, and higher amount of genes coding for enzymes involved in the hemebiosynthetic pathway respect to wild type plants, KD501 and KD681 showed theopposite phenotype. The expression of genes coding for catalase was not altered in allthe lines. However, catalase activity in the mitochondrial fraction of the mutant linescould be restored after incubation with hemin. The expression of genes that code for FHand other Fe-S cluster assembly proteins were also increased in MycA and MycB anddecreased in KD501 and KD681. These results suggest that there is a close relationshipbetween the Fe-S cluster assembly and the heme biosynthetic pathway, and we suggestthat FH could be the link between them.
