Resumen:
Development of the human placenta is critical for a successfulpregnancy. It requires that the extravillous cytotrophoblast cells(EVTs) migrate, invade and transform maternal spiral arteries. Itwas recently reported that aquaporin 3 (AQP3) is expressed in trophoblastcells during early gestation and its expression decreaseddramatically in preeclamptic placentas. Although, AQP3 is involvedin the migration of different cell types, its role in human placenta isstill unknown.Objective: To evaluate the role of AQP3 in the migration and theinvasion of EVT.Methods: Trophoblast cells (Swan 71 cell line) were cultured incomplete DMEM-F12. AQP3 expression was examined by Westernblot, RT-PCR and immunofluorescence. Cells were also treated with100 μM CuSO4 (AQP3 specific blocker) or SiRNA-AQP3 to inhibitAQP3. Migration was assessed by wound healing assay. Transwellinvasion assay was also performed. Metalloproteinase activitieswere evaluated by gelatin gel zymography.Results: AQP3 expression was found in Swan-71.
