NEW INSIGHTS INTO THE CROSSTALK BETWEEN PLASTID AND MITOCHONDRIAL RETROGRADE SIGNALLING PATHWAYS.

My scientific goal is to delve into theinteractions between retrograde signaling pathways in plants. To get this goal,I have been interested in resolving the intracellular localisation of Kinase1and address it to its function. Using N.benthamiana to transiently express the main splicing form of the kinasefused to eGFP driven by its own promoter, we identified its presence in both nucleiand endoplasmic reticulum (ER). Interestingly, when tissues expressing thefluorescent construct were treated with compounds perturbing chloroplaststatus, drastic changes in the Kinase1 distribution between ER and nucleus wereobserved suggesting an ER-organelles communication that regulates Kinase1intracellular dynamic. In order to ascertain this dual localisation, thesensing of organelles status and its dynamics as secondary messenger in Kinase1-dependentsignaling pathways, I am conducting analysis by confocal microscopy of Arabidopsistransgenic stable lines and starting to add to my model experiments in hepaticcancer cells (HCC from HepG2/C3A and HuH-7) and Danio rerio to takeadvantage of the conservation of signaling cellular pathways. In addition, Iuse Arabidopsis mutant backgrounds in cellular and mitochondrial energyhomeostasis to reveal connections between the intracellular Kinase1 localisationand its function as central integrator of energy imbalance signals modifyingnuclear gene expression.
