Resumen:
The human protein Glrx3, also known as PICOT(PKC Interacting Cousin of Thioredoxin), has been related with many humandiseases such as lung and colon cancer, Hodgkin?s lymphoma, diseases related toiron misregulation and to the immune response. Glrx3 presents homology withGrx3 and Grx4 glutaredoxins of budding yeast. The double mutant grx3grx4displays an important impairment in the Chronological Life Span (CLS) comparedto wild type cells. We have cloned Glrx3 in a yeast vector and have observedthat is capable to restore life span in the grx3grx4 double mutantconcomitantly with a mild restoration of Slt2 activity at late points of the CLSexperiment. Slt2 also restores life span in the simultaneous absence ofGrx3/Grx4. Interestingly, Glrx3 partly complements the defects in lifeextension caused by the absence of Slt2. We have identified a new function forGrx3/Grx4 in CLS dependent on the PKC1-MAPK pathway. Glrx3 presents a highdegree of complementation for this function in the humanized yeast, suggestingan elevated degree of conservation in human cells that can be extraordinaryuseful for future research.
