High throughput sequencing of Lotus corniculatus L. and Lotus tenuis Waldst. Et Kit

Previous studies carried out in our laboratory suggests that L. tenuis (Lt) and L. corniculatus (Lc)are an invaluable tool to study the genetic control of proanthocyanidins (PAs) biosynthesis andtolerance to stresses in legumes. With these aim we performed an RNA-Seq sequencing to obtainthe full-length transcriptome of both species. For these, RNA samples were isolated from shootsof a diploid accession of Lc and Lt (n=3) and were used to performed a high-performance paired-end (2x100bp) sequencing on Hiseq 1500-Illumina. From obtained reads we performed twodifferent analyses: a) de novo assembly using Trinity program and b) assembling and mapping toL. japonicus genome using TopHat and Cufflinks programs. From a) analysis, we obtained91,530 and 80,911 genes for Lc and Lt respectively, functional annotation of these genes wasperformed using Trinotate pipeline. On the other hand, between 56 to 59% of transcripts weremapped against L. japonicus genome from b) analysis; allowing the identification of 42,013genes. From these, 12,868 genes were differentially expressed between Lc and Lt, and 4,912 ofthose had fold change ≥|2|. GO enrichment analysis by Blast2GO of differentially expressedgenes was performed to determinate the GO terms highly represented. This work represents the first report of full transcriptome of L. tenuis and a diploid L. corniculatus, raising the possibilityof future agricultural studies and applications in the others forage legumes.0575
