Dehydroleucodine Inhibits the Proliferation of Mouse Melanoma Cells

Dehydroleucodine (DhL) is a sesquiterpene lactone extracted from the medicinal plant Artemisia douglasiana. Results of our laboratory showed that DhL delays the proliferation of HeLa cells, arresting them in the G2 phase of the cell cycle. In this work we proposed to analyze whether DhL inhibits the proliferation of B16-F0 and B16-F10 cells and alters cell cycle proteins. B16 F0 y B16 F10 cells were obtained from a spontaneous mouse C57BL/6 melanoma. 1x104 quiescent cells were stimulated with 10% of fetal bovine serum (SFB) in the presence of 0-6 µM DhL by 72 h. The growth index was determined (IC ± ESM) each 24 h and the results were analyzed by the variance test. Proteins of the cell cycle were analyzed by western blot. Results: After 72 h of culture control B16 F0 and B 16 F10 cells showed an IC of 9,7 ± 0,3 and 10,5 ± 1,1 respectively, with 2 µM DhL 5,7 ± 0,5 and 6,4 ± 0,7 respectively, with 4 µM 1,7 ± 0,3 and 1,9 ± 0,1 respectively, with 6 µM 1,0 ± 0,1 and 1,4 ± 0,3 respectively. Statistical analysis showed significant difference among 0, 2, 4 and 6 µM DhL. Also, incubation with 4 y 6 µM DhL reduced the concentration of ciclin B1 and survivin. DhL inhibits the proliferation of the B16 F0 and B16 F10 cells in a concentration dependent way, decreasing the expression of ciclin B1 and survivin.
