Fear response throughout age and life history: behavioral characterization of the effects of adolescent stress on a fear conditioning paradigm in Wistar rats

The goal of this study was to evaluate the behavioral response to contextual fear conditioning paradigm (CFCP), hypothesizing that fear responses along life are enhanced when facing chronic variable stress during adolescence, a period known to be highly vulnerable to stress. Rats PND40 were subject to chronic variable stress (CVS) for 2 weeks: 5 stressors were presented randomly twice daily and twice a week 2 overnight stressors. Controls underwent only normal animal handling. A group was allowed to recovery 1 week before being submitted to a CFCP (short-term group ) starting at PND 61. Other group was tested 5 weeks after CVS (long-term group) PND 90. CFCP: training (3-min exploration- 3 shocks 1mA, 1s, 1min apart - 3 min), 4 days extinction (5 min) and recall 48 hs extinction (1-min,1 shock: 1 mA, 1s, 3-min). Behavioral parameters: freezing, grooming, horizontal and vertical activity. Age effects: The analysis showed that in the different phases of the test, all the behavioral parameters vary along life.
