LECTINHISTOCHEMICAL STUDY OF THE OESOPHAGUS, PROVENTRICULUS AND GIZZARD OF THE GALLARETA CHICA (Fulica leucoptera)

Using the PAS histochemical technique we have previously shown that epithelial cells of Fulica leucoptera digestive tract present glycoconjugate components. The aim of the present study was to characterize those glycoproteins using a lectinhistochemical technique. Oesophagus and stomachs samples of three animals were embedded in paraffin and incubated with the following biotinylated lectins: Con-A, SBA, DBA, RCA-1, PNA, UEA-1, and WGA. Steptavidin-Biotin was the detection system, 3,3´diaminobenzidine tetrahydrocloride the chromogen and Mayer´s hematoxylin the counterstain. SBA strongly bound to all the surface epithelial glycocalix, and to gizzard and proventriculus glandular epithelia. WGA, PNA y RCA-1 intensely bound to oesophagic epithelium, surface epithelia glycocalix and deep proventricular ducts. RCA-1 and Con-A labelled cells at the base of the superficial proventricular glands. Gizzard did not react to DBA, WGA or PNA. UEA-1 labelling was only present in the most superficial epithelial layers of the oesophagus. From the results we can conclude that studied epithelia have little amount of a-L fucosa residues; ß-D-GlcNAc; NeuNAc, ß-D-Gal(ß1-3)>D-GalNAc, a-D-GalNAc are not present in gizzard epithelia and a-D-GalNAc; ß-D-GalNAc can be found in all the analysed structures.
