VIP induces the decidualization program on human endometrial stromal cells and conditions dendritic cells

The decidualization process is critical to achieve a successful embryo implantation. While progesteron is a key hormone within this process, it is triggered by an intracitoplasmatic increase of cAMP in the stromal endometrial cells. The Vasoactive Intestinal Peptide (VIP) is a neuropeptide with immunoregulatory activity produced by endometrial, trophoblastic and immune cells among others, and its main signalling pathway involves cAMP. Previously, we reported that VIP mediates tolerogenic e?ects in the mateno-placental interface.ObjectiveInvestigate VIP contribution in the decidualization process through an in vitro di?erentitaion and implantation model and its e?ect on monocytes and dendritic cells.ConclusionVIP contributed to the decidualization process by inducing characteristic markers of decidualization and of endometrial receptivity. Also, from a funtional point of view, in our in vitro model, VIP decidualized HESC cells allowed trophoblast cell invasion and induced a tolerogenic prole on dendritic cells, as observed with MPA-cAMP decidualized cells.These results suggest that VIP could contribute to the decidualization process on human endometrial stromal cells.
