Resumen:
Erythrocytes may be susceptible to peroxidation by a high amount of polyunsaturated fatty acids, exposure to high concentrations of oxygen and having transition metals that can act as catalysts. The aim of this study is to evaluate the peroxidative damage on erythrocytes by chemiluminescence initiated by tert-butyl hydroperoxide. We worked with blood from: a) human pathologies: sepsis, alcoholism and heart; b) Experimental Models: sepsis, alcoholism, pancreatitis and pseudosaturnismo. Heparinized blood was washed 3 times with 150 mM NaCl being 75/500 working dilution was used. Reading the buffer was 120 mM NaCl, 30 mM phosphate. The determination was carried out in a liquid scintillation counter with circuit switched coincidence. To initiate the reaction was added tert-butyl hydroperoxide (3 mM). The results showed that in human pathologies there is an increase in the emission of 55 and 53% for sepsis and alcoholism respectively and a decrease of 33% for coronary (VC: 8.14 ± 0.06 x 10 3 cpm / mg hemoglobin). 0.10 ± 8.7 x: as in experimental models, emission increases reaching 43% in sepsis, in pseudosaturnismo 26%, 12% in pancreatitis, alcoholism in 85% (VC were observed 10 3 cpm / mg hemoglobin) .These results suggest that this technique could be used for monitoring oxidative stress in different pathologies
