EFFECT OF LONG-TERM ADMINISTRATION OF ENALAPRIL MALEATE ON THE PLOIDY STATUS OF MURINE HEART

One of the proposed mechanisms
for cardiac remodeling is myocyte replication. On the basis that inhibitors of angiotensin
converting enzyme (ACEI) are able to influence heart remodeling we decided to
investigate if the prolonged administration of an ACEI, enalapril maleate (EM),
is able to modify the DNA content of the heart muscle cells nuclei in normal
mice. Female 3 month-old BALB/c mice received for 50 and 150 days EM in the
drinking water (300mg/L). Controls were mice of matching age and sex. In
Feulgen stained tissue sections of the left ventricle free wall the DNA content
of individual myocyte nuclei was determined by densitometry with a digital
analysis system. In animals receiving ACEI for 50 days (n=ll) 5.6% ± 0.75 (SEM)
of myocyte nuclei were tetraploid as compared with 2.1%±0.46 in control mice
(n=6) (p<0.04). In 6 mice treated for 150 days with ACEI the number of
tetraploid myocyte nuclei was significantly higher (p<0.03) than in 6
control non-treated mice (9.66±1.06 and 2±0.48 respectively). These results, showing
that the prolonged treatment of normal mice with an ACEI increases the number
of tetraploid myocytes, suggest that ACEI may particípate in remodeling by
production of polyploid myocytes either by induction of DNA synthesis, by nuclear
fusión or by preventing división of tetraploid nuclei.
